Bruce Banner became the Hulk in a gamma radiation experiment run by General Thaddeus Ross. On the run, he hides in Rio de Janeiro, works at a soda bottling plant and controls his heart rate to avoid transforming. He seeks a cure with an online contact, "Mr. Blue." A drop of his blood in a soda leads Ross to him. Soldier Emil Blonsky gets a version of the serum and becomes obsessed with the Hulk. Banner reunites with Betty Ross, the general's daughter and his old love. Mr. Blue is scientist Samuel Sterns. Blonsky forces Sterns to give him Banner's blood and becomes the Abomination, tearing up Harlem. The Hulk beats him and spares him. In the end, Tony Stark tells Ross a team is being put together.

ReferenceBill Bixby on Bruce's TV
Early on, in Rio, Bruce watches a clip of The Courtship of Eddie's Father. The actor on screen is Bill Bixby, the Bruce Banner of the '70s TV series.
The security guard who lets Bruce into the university in exchange for pizza is Lou Ferrigno, the Hulk from the classic series. He also voices the monster in the movie.
Bruce tries to threaten the thugs at the Rio factory in clumsy Portuguese and ends up saying they won't like him when he's hungry. It was supposed to be "angry," the classic line from the TV series.
Near the end, Bruce's blood drips into a cut on Samuel Sterns' head and he starts to change. That's the origin of the villain the Leader in the comics, and the character comes back later in the franchise.
